PVT1: A Cancer-associated Non-coding Gene Revisited

چکیده

PVT1 was originally identified as a transcriptional unit from a human homologous sequence to Pvt1, which was cloned from murine plasmacytoma with t(6;15). Previous studies have revealed various genetic alterations in the PVT1 locus, including chromosome translocation, amplification, chromothripsis, and single nucleotide polymorphisms in human diseases, suggesting important roles of PVT1 in the pathogenesis. However, because this locus does not produce protein coding sequences, its functional properties have not been characterized and its biological significance remains unclear. Recent studies have shown that the PVT1 locus encodes lincRNAs and microRNAs. Therefore, current investigations are being performed focusing on the biological features of this long-standing puzzle gene as a non-coding gene. *Corresponding author: Takashi Sonoki, Hematology/Oncology, Wakayama Medical University, 811-1 Kimi-idera, Wakayama, 641-8510, Japan, Tel: +81-73441-0665; Fax: +81-73-441-0653; E-mail: [email protected] Received August 24, 2013; Accepted January 15, 2014; Published January 17, 2014 Citation: Sonoki T (2014) PVT1: A Cancer-associated Non-coding Gene Revisited. Induction of apoptosis and necrosis in human acute erythroleukemia cells by inhibition of long non-coding RNA PVT1

Recent advances in molecular medicine have proposed new therapeutic strategies for cancer. One of the molecular research lines for the diagnosis and treatment of cancer is the use of long non-coding RNAs (LncRNAs) which are a class of non-coding RNA molecules longer than 200 base pairs in length that act as the key regulator of gene expression. Different aspects of cellular activities like cell...

Long non-coding RNA PVT1 is associated with tumor progression and predicts recurrence in hepatocellular carcinoma patients

PVT1, which maps to chromosome 8q24, is a copy number amplification-associated long non-coding RNA. Overexpression of PVT1 is a powerful predictor of tumor progression and patient survival in a diverse range of cancer types. However, the association between PVT1 and hepatocellular carcinoma (HCC) remains unclear. The aim of the present study was to examine the expression pattern of PVT1, and it...

PVT1‐derived miR‐1207‐5p promotes breast cancer cell growth by targeting STAT6

Accumulating evidence indicates that ectopic expression of non-coding RNAs are responsible for breast cancer progression. Increased non-coding RNA PVT1, the host gene of microRNA-1207-5p (miR-1207-5p), has been associated with breast cancer proliferation. However, how PVT1 functions in breast cancer is still not clear. In this study, we show a PVT1-derived microRNA, miR-1207-5p, that promotes t...

LncRNA plasmacytoma variant translocation 1 is an oncogene in bladder urothelial carcinoma

Bladder cancer (BC) is the most lethal malignant cancer of the genitourinary system, and bladder urothelial carcinoma (BUC) is the most common type of BC. The long non-coding RNA (lncRNA) plasmacytoma variant translocation 1 (PVT1) is overexpressed in several malignant tumors, including BC. Using a lncRNA array and quantitative real-time PCR, we detected greater expression of PVT1 in BUC tissue...

High expression of lncRNA PVT1 promotes invasion by inducing epithelial-to-mesenchymal transition in esophageal cancer

The long non-coding RNA (lncRNA) plasmacytoma variant translocation 1 (PVT1) has been identified as an oncogene in numerous diseases, and aberrant lncRNA PVT1 expression has been associated with the development of cancer. However, the underlying mechanism by which lncRNA PVT1 affects cell invasion in esophageal cancer has been not demonstrated. In the current study, the expression of lncRNA PVT...
